چکیده

Tongue squamous cell carcinoma (TSCC) is an aggressive neoplasm which typically spreads through invasive growth with serious morbidity and mortality. Previous studies suggested that the expression of LRIG1 functions as the vital suppressor gene in various human cancer types by inhibiting invasion and proliferation. However, little is known about the downstream genes coordinating this process in tongue squamous cell carcinoma. In this study, we overexpressed LRIG1 with a full-length expression vector and used small interfering RNA (siRNA) to block LRIG1 gene in the Tca8113 TSCC cell line. The expression of LRIG1 and EGFR at mRNA and protein level were tested by qRT-PCR and Western blot, respectively. The transwell assay and CCK8 assay were performed to detect the invasion and proliferation of TSCC cells. LRIG1 overexpression reduced EGFR expression, and significantly inhibited the invasion and proliferation of TSCC cells. In addition, silence of endogenous LRIG1 expression resulted in elevated EGFR expression and increased cell invasion and proliferation. Our result suggested that LRIG1 restricted TSCC invasion and proliferation via regulating the EGFR pathway. Restoration of LRIG1 in TSCC cells could be used as a novel therapeutic strategy.

LRIG1 regulates cadherin-dependent contact inhibition directing epithelial homeostasis and pre-invasive squamous cell carcinoma development

Epidermal growth factor receptor (EGFR) pathway activation is a frequent event in human carcinomas. Mutations in EGFR itself are, however, rare, and the mechanisms regulating EGFR activation remain elusive. Leucine-rich immunoglobulin repeats-1 (LRIG1), an inhibitor of EGFR activity, is one of four genes identified that predict patient survival across solid tumour types including breast, lung, ...

Overexpression of LRIG1 regulates PTEN via MAPK/MEK signaling pathway in esophageal squamous cell carcinoma

The present study aimed to evaluate the role of leucine-rich repeats and immunoglobulin-like domain protein 1 (LRIG1) in the regulation of phosphatase and tensin homolog (PTEN) expression in esophageal carcinogenesis. LRIG1 was overexpressed in esophageal squamous cell carcinoma (ESCC) cell lines, and the effect of LRIG1 overexpression on the mRNA and protein expression levels of PTEN was evalu...

MicroRNA-509 acts as a tumor suppressor in tongue squamous cell carcinoma by targeting epidermal growth factor receptor

Tongue squamous cell carcinoma (TSCC) is the most frequent type of oral carcinoma, and is characterized by high metastatic and growth capabilities. Previous studies have demonstrated that aberrantly expressed cancer‑associated microRNAs (miRs) may be associated with tumorigenesis and tumor development in various types of cancer, including TSCC. miR‑509 has been identified as a critical regulato...

A novel treatment approach for retinoblastoma by targeting epithelial growth factor receptor expression with a shRNA lentiviral system

Objective(s): Non-invasive treatment options for retinoblastoma (RB), the most common malignant eye tumor among children, are lacking. Epithelial growth factor receptor (EGFR) accelerates cell proliferation, survival, and invasion of many tumors including RB. However, RB treatment by targeting EGFR has not yet been researched. In the current study, we investigated the effect of EGFR down-regula...

Effect of A disintegrin and metalloproteinase 10 gene silencing on the proliferation, invasion and migration of the human tongue squamous cell carcinoma cell line TCA8113

The present study aimed to investigate the effect of A disintegrin and metalloproteinase 10 (ADAM10) gene silencing on the proliferation, migration and invasion of the human tongue squamous cell carcinoma cell line TCA8113. RNA interference was used to knock down the expression of ADAM10 in the TCA8113 cell line and the proliferation, migration and invasive ability of the treated cells were obs...
